Increased thiamine intake may be required to maintain thiamine status during weight loss in patients with type 2 diabetes
Brinkworth, GD; Noakes, M; Cleanthous, X; Wycherley, TP; Clifton, PM; Keogh, JB
DIABETES RES CLIN PRACT - 01 Dec 2012
During weight loss, erythrocyte thiamine pyrophosphate (TPP) decreased (221±52 to 195±39 nmol/L, P<0.05) on a diet with adequate thiamine (1.1 mg/day) but was unchanged (217±55 vs 218±52 nmol/L, NS) on a high thiamine diet (2.8 mg/day). Attention to thiamine status may be required in patients with diabetes after weight loss. Copyright © 2012 Elsevier Ireland Ltd. All rights reserved.
The metabolic syndrome and cancer: is the metabolic syndrome useful for predicting cancer risk above and beyond its individual components?
Davis, TM; Davis, WA; Sooriyakumaran, M; Dobson, A; Pasco, JA; Tonkin, A; Harding, J; Simmons, D; Knuiman, M; Shaw, JE; Mitchell, P; Woodward, M; Magliano, DJ; Anstey, KJ; Grant, J; Briffa, T; Luszcz, M; Balkau, B; Adams, R; Giles, GG; Reid, C; Simons, L
DIABETES METAB - 01 Dec 2015
AIMS: The metabolic syndrome (MetS) is a risk factor for cancer. However, it is not known if the MetS confers a greater cancer risk than the sum of its individual components, which components drive the association, or if the MetS predicts future cancer risk. MATERIALS AND METHODS: We linked 20,648 participants from the Australian and New Zealand Diabetes and Cancer Collaboration with complet...
Regulation and actions of activin A and follistatin in myocardial ischaemia-reperfusion injury
Chen, Y; Kaye, D; Smith, JA; Verrier, E; Philips, DJ; Venardos, K; Hedger, MP; Spring, D; Rothnie, C
CYTOKINE - 01 Oct 2014
Activin A, a member of the transforming growth factor-β superfamily, is stimulated early in inflammation via the Toll-like receptor (TLR) 4 signalling pathway, which is also activated in myocardial ischaemia-reperfusion. Neutralising activin A by treatment with the activin-binding protein, follistatin, reduces inflammation and mortality in several disease models. This study assesses the regulat...

Galam, DL; Siddique, MM; Meikle, PJ; Gopalan, V; Sigmundsson, K; Kaddai, VA; Weir, JM; Sugii, S; Summers, SA; Henstridge, DC; Yuguang, G; Mellet, NA; Velan, SS; Sriram, S; Wang, M; Bin M Yassin, MS; Tsong, TJ; Shabbir, A; Chaurasia, B; Shayman, JA; Hirabayashi, Y; Bulchand, S; Shiow, ST; Prakash, KN; Lancaster, GI
CELL METAB - 13 Dec 2016
Adipocytes package incoming fatty acids into triglycerides and other glycerolipids, with only a fraction spilling into a parallel biosynthetic pathway that produces sphingolipids. Herein, we demonstrate that subcutaneous adipose tissue of type 2 diabetics contains considerably more sphingolipids than non-diabetic, BMI-matched counterparts. Whole-body and adipose tissue-specific inhibition/delet...

Dragoljevic, D; Cengia, L; Vince, JE; Masters, SL; Lane, RM; Gerlic, M; Harrison, LC; Murphy, AJ; Lee, MK; Whitehead, LW; Kammoun, HL; Kile, BT; Vasanthakumar, A; Croker, BA; DiRago, L; Kraakman, MJ; Lawlor, KE; Metcalf, D; Febbraio, MA; Kallies, A; Wentworth, JM
CELL METAB - 12 Jan 2016
Interleukin-18 (IL-18) is activated by Caspase-1 in inflammasome complexes and has anti-obesity effects; however, it is not known which inflammasome regulates this process. We found that mice lacking the NLRP1 inflammasome phenocopy mice lacking IL-18, with spontaneous obesity due to intrinsic lipid accumulation. This is exacerbated when the mice are fed a high-fat diet (HFD) or a high-protein ...
Fetuin B is a secreted hepatocyte factor linking steatosis to impaired glucose metabolism
Kraakman, MJ; Febbraio, MA; Meex, RC; Rensen, SS; Goode, RJ; Lo, JC; Morris, A; Burke, M; Hoy, AJ; Molloy, MP; Bruce, CR; Watt, MJ; Kingwell, BA; Brown, RD; Greve, JW; Lancaster, GI
CELL METAB - 01 Dec 2015
Liver steatosis is associated with the development of insulin resistance and the pathogenesis of type 2 diabetes. We tested the hypothesis that protein signals originating from steatotic hepatocytes communicate with other cells to modulate metabolic phenotypes. We show that the secreted factors from steatotic hepatocytes induce pro-inflammatory signaling and insulin resistance in cultured cells...
The CDP-ethanolamine pathway regulates skeletal muscle diacylglycerol content and mitochondrial biogenesis without altering insulin sensitivity
Genders, AJ; Jackowski, S; Febbraio, MA; Lee-Young, RS; Lamon, S; Risis, S; Meikle, PJ; McGee, SL; Selathurai, A; Sepulveda, P; Frank, M; Kowalski, GM; Bruce, CR; Watt, MJ; Burch, ML; Russell, AP
CELL METAB - 05 May 2015
Accumulation of diacylglycerol (DG) in muscle is thought to cause insulin resistance. DG is a precursor for phospholipids, thus phospholipid synthesis could be involved in regulating muscle DG. Little is known about the interaction between phospholipid and DG in muscle; therefore, we examined whether disrupting muscle phospholipid synthesis, specifically phosphatidylethanolamine (PtdEtn), would...
Blocking IL-6 trans-signaling prevents high-fat diet-induced adipose tissue macrophage recruitment but does not improve insulin resistance
Febbraio, MA; Yang, C; Lee-Young, RS; Allen, TL; Neill, B; Murphy, AJ; Du, XJ; Kammoun, HL; Henstridge, DC; Bobik, A; White, DA; Kingwell, BA; Kallies, A; Deswaerte, V; Vasanthakumar, A; Risis, S; Kraakman, MJ; Estevez, E; Shi, W; Peijs, L; Matthews, VB; Rose-John, S; Bruce, CR; Lancaster, GI
CELL METAB - 03 Mar 2015
Interleukin-6 (IL-6) plays a paradoxical role in inflammation and metabolism. The pro-inflammatory effects of IL-6 are mediated via IL-6 "trans-signaling," a process where the soluble form of the IL-6 receptor (sIL-6R) binds IL-6 and activates signaling in inflammatory cells that express the gp130 but not the IL-6 receptor. Here we show that trans-signaling recruits macrophages into adipose tis...

Gorman, DJ; Choi, SH; Goldberg, IJ; Nagareddy, PR; Hong, ES; Smyth, SS; Grant, RW; Kraakman, M; Fisher, EA; Murphy, AJ; Bornfeldt, KE; Korner, J; Masters, SL; Dixit, VD; Abdel-Latif, A; Dragoljevic, D; Stirzaker, RA; Tall, AR
CELL METAB - 06 May 2014
Obesity is associated with infiltration of macrophages into adipose tissue (AT), contributing to insulin resistance and diabetes. However, relatively little is known regarding the origin of AT macrophages (ATMs). We discovered that murine models of obesity have prominent monocytosis and neutrophilia, associated with proliferation and expansion of bone marrow (BM) myeloid progenitors. AT transpl...
ABCA12 regulates ABCA1-dependent cholesterol efflux from macrophages and the development of atherosclerosis
Smyth, IM; Henley, KJ; Ditommaso, K; Jones, L; Sviridov, D; D'Souza, W; Mukhamedova, N; Kesani, R; Lane, RM; Fu, Y; Ditiatkovski, M; Kile, BT; Jennings, G; Ip, S
CELL METAB - 06 Aug 2013
ABCA12 is involved in the transport of ceramides in skin, but it may play a wider role in lipid metabolism. We show that, in Abca12-deficient macrophages, cholesterol efflux failed to respond to activation with LXR agonists. Abca12 deficiency caused a reduction in the abundance of Abca1, Abcg1, and Lxrβ. Overexpression of Lxrβ reversed the effects. Mechanistically, Abca12 deficiency did not aff...

Wu, H; Zhang, Y; Lan, B; Pei, M; Fan, F; Dart, AM; Du, Y; Zou, Y; Song, Q; Zhang, S; He, L
CLIN THER - 01 Sep 2017
PURPOSE: Peripherally inserted central catheters (PICCs) are frequently used for prolonged drug administration, but their use is commonly complicated by the development of upper extremity deep venous thrombosis (UEDVT) requiring anticoagulation. Here, we compared the efficacy and safety profile of rivaroxaban (20 mg/d) with low molecular weight (LMW) heparin and vitamin K antagonists in the ...
Comparison of creatinine and cystatin C based eGFR in the estimation of glomerular filtration rate in Indigenous Australians: the eGFR Study
Maple-Brown, LJ; Jerums, G; Sajiv, C; Barr, EL; Hughes, JT; Barzi, F; Ekinci, EI; Brown, AD; Hoy, WE; Cass, A; Jones, GR; Lawton, PD; Ellis, AG; O'Dea, K; MacIsaac, RJ; Majoni, SW
CLIN BIOCHEM - 01 Apr 2017
BACKGROUND: The Chronic Kidney Disease Epidemiology Collaboration (CKD-EPI) equation that combines creatinine and cystatin C is superior to equations that include either measure alone in estimating glomerular filtration rate (GFR). However, whether cystatin C can provide any additional benefits in estimating GFR for Indigenous Australians, a population at high risk of end-stage kidney diseas...

Bond, ST; Bruce, CR; McGee, SL; McEwen, KA; Ashton, TD; Fleming, C; Henstridge, DC; Chen, D; Walder, KR; Gregorevic, P; Martin, SD; Hargreaves, M; Connor, T; Sanigorski, A; Wu, M; Kerr-Bayles, L; Gaur, V; Pfeffer, FM; Febbraio, MA; Schwabe, JW; Baar, K; Hudson, GM; Pike Winer, LS
CELL REP - 13 Sep 2016
Drugs that recapitulate aspects of the exercise adaptive response have the potential to provide better treatment for diseases associated with physical inactivity. We previously observed reduced skeletal muscle class IIa HDAC (histone deacetylase) transcriptional repressive activity during exercise. Here, we find that exercise-like adaptations are induced by skeletal muscle expression of class I...
Cytomegalovirus restructures lipid rafts via a US28/CDC42-mediated pathway, enhancing cholesterol efflux from host cells
Religa, P; Liu, Y; Ditiatkovski, M; Low, H; Meikle, PJ; Fu, Y; Avdic, S; McSharry, BP; Mukhamedova, N; Blomberg, M; Hoang, A; Miller, WE; Polyzos, KA; Sviridov, D; Slobedman, B; Soderberg-Naucler, C; Bukrinsky, M; Cui, HL
CELL REP - 28 Jun 2016
Cytomegalovirus (HCMV) contains cholesterol, but how HCMV interacts with host cholesterol metabolism is unknown. We found that, in human fibroblasts, HCMV infection increased the efflux of cellular cholesterol, despite reducing the abundance of ABCA1. Mechanistically, viral protein US28 was acting through CDC42, rearranging actin microfilaments, causing association of actin with lipid rafts, an...
Zebrafish embryonic lipidomic analysis reveals that the yolk cell is metabolically active in processing lipid
Sanigorski, A; Gibert, Y; Meikle, PJ; Sinclair, AJ; Mellett, NA; Fraher, D
CELL REP - 16 Feb 2016
The role of lipids in providing energy and structural cellular components during vertebrate development is poorly understood. To elucidate these roles further, we visualized lipid deposition and examined expression of key lipid-regulating genes during zebrafish embryogenesis. We also conducted a semiquantitative analysis of lipidomic composition using liquid chromatography (LC)-mass spectrometr...

Lynch, GS; Anderton, H; Silke, J; Laine, D; Casey, JL; Tanzer, M; Schloegel, J; Sahay, T; Dillon, H; Weston, R; Liu, SY; Wong, WW; Talbo, G; Schneider, P; Scott, AM; Hoogenraad, J; Gregorevic, P; Foley, M; Jayatilleke, KM; Johnston, AJ; Faou, P; Emmrich, K; Hoogenraad, NJ; Levina, V; Burkly, LC; Jenkinson, L; Murphy, KT; Hall, C
CELL - 10 Sep 2015
The cytokine TWEAK and its cognate receptor Fn14 are members of the TNF/TNFR superfamily and are upregulated in tumors. We found that Fn14, when expressed in tumors, causes cachexia and that antibodies against Fn14 dramatically extended lifespan by inhibiting tumor-induced weight loss although having only moderate inhibitory effects on tumor growth. Anti-Fn14 antibodies prevented tumor-induced ...
